Orsi Szecsi led the Dukes with 20 points. Wumi Agunbiade scored 13 and Samantha Pollino added 11 for the Dukes who fell to 22-8 overall.
Justine Raterman led all scorers with 21 points, Kristin Daugherty finished with 18 and was 4-of-5 from beyond the arc and Patrice Lalor added 10 for the Flyers who improve to 20-10.
The Dukes started out cold from the floor, hitting only 3 of its first 18 shots while falling behind 17-7 midway through the first half.
With UD leading 23-13, the Dukes went on a 9-0 run capped by an Abel pull-up jumper at the 2:27 mark.
However, the Flyers would hold the Dukes scoreless the rest of the half as UD scored the next seven points to take a 30-22 lead into halftime.
Down nine points at the 17:35 mark of the second half, Duquesne would close the gap to one point over next minute and a half making the score 37-36.
A Szecsi 3-pointer followed by a Pollino foul shot tied the contest at 40-40 with 14:09 to play, but Flyers answered right away with a Daugherty 3-pointer.
The teams would play even over the next six minutes, but Duquesne would only hit three field goals in the final eight minutes as the Flyers would pull away down the stretch.
